体外研究 | KX2-391是一种Src抑制剂,直接作用于Src底物袋。KX2-391对Huh7 (GI 50 = 9 nM),PLC/PRF/5 (GI 50 = 13 nM),Hep3B (GI 50 = 26 nM),和HepG2 (GI 50 = 60 nM),四种肝癌(HCC)细胞系表现出陡峭的剂量反应曲线。 KX2-391能够抑制某些对市售药物耐药的白血病细胞,比如衍生自负荷T3151突变体的慢性白血病细胞的细胞系。KX2-391在设计的Src驱动细胞生长试验中进行评估,在NIH3T3/c-Src527F和SYF/c-Src527F细胞中的GI50分别为23 nM 和39 nM。 |
